The land where Villanova was established originally belonged to the Lenni Lenape Indians but was settled by Quakers from Radnorshire between 1663-1665. The group was seeking religious freedom and purchased the land from William Penn, who had acquired it through a grant from the Crown. The official founding of Radnor Township occurred in 1682 when it was deemed Penn's Green Country.

During the Revolutionary War, it was significantly uninhabited and hosted a few battles between the British and the Continental Army. Eventually, in 1794, the first toll road in America was built through it and named it the Lancaster Turnpike, which today is known as Route 30.

In 1842, Villanova University was founded in the area and dubbed St. Thomas of Villanova College.

Villanova is a wealthy area along the Main Line of Philadelphia, which originated as an extension of the Pennsylvania Railroad, connecting towns west of the city, and facilitating the commute of rich industrialists from their work in the city to their country homes, gentleman clubs, and summer residences.
